Brief description of the Centre/Research Group (including URL if applicable):

The Institute of Catalysis and Petrochemistry (ICP) is a research center belonging to the Spanish National Research Council (CSIC), which is the largest public institution dedicated to research in Spain. ICP’s main purpose is to develop new technological solutions within the field of catalysis, which contribute to improving the quality of life of citizens and enhance the competitiveness of our chemical industry, within the concept of Green Chemistry. Its research is oriented towards the development of advanced catalysts, both inorganic and enzymatic ones, along three main lines: Energy, Environmental Protection and Selective Synthesis of Chemical Compounds.

The Bioelectrocatalysis Group of the ICP has as scientific objectives of the group the functional/structural characterization of metalloenzymes with redox properties and their interaction with electrodes for applications in biosensors,biofuel cells, bioelectronsynthesis and photobioelectrocatalysis.   For this purpose we have focused in the last years in the preparation and characterization of nano-structured electrodes as a platform for controlled biocatalyst immobilization using gold nanoparticles, carbon nanotubes, graphene, semiconductor particles or biomimetic membrane bilayers. The group is formed by two staff scientists and a variable number of postdoctoral and predoctoral researchers. In the last 5 years the group has participated in 3 national research projects, 3 european ones and 1 contract with an industrial company.

Project description:

The project aims the reconstitution of an ATPase, a membrane-bound enzyme that produces/consumes ATP, on biomimetic membranes supported on gold electrodes. ATP, the molecule used by living organisms to supply energy to many different metabolic processes, is synthesized mostly by the ATPase synthase using a proton or sodium gradient generated across a lipid membrane. Modified electrode surfaces will be prepared to integrate a hydrogenase, a  redox enzyme that catalyzes hydrogen oxidation, and a ATPase in a supported phospholipid bilayer.  The electroenzymatic oxidation of H2 will be coupled to the synthesis of ATP. Proton gradients generated across the biomimetic membrane will be studied electrochemically and the production of ATP from ADP and phosphate will be measured. The goal will be to design a bionanoelectronic device that can regenerate ATP using hydrogen as energy source. Finally the ATP regeneration system will be studied for several enzyme-catalyzed phosphorylation and carboxylation reactions.

The following techniques will be used to study the system: electrochemical methods, atomic force microscopy (AFM), quartz crystal microbalance (QCM), high performance liquid chromatography (HPLC), UV-vis spectroscopy.
